Akt1, Akt2, and Akt3 make up a family of serine/threonine kinases, often activated downstream of growth factor receptors and PI 3-Kinase. Each Akt family member contains an N-terminal pleckstrin homology (PH) domain, a central kinase domain, and a C-terminal regulatory domain.
Main roles are inhibition of apoptosis and promotion of proliferation. AKT has been shown to play a role in such metabolic processes as glucose transport, glycogen synthesis, glycolysis, and protein synthesis. Akt perturbations are thought to play an important role in tumorigenesis.
Image: rhAkt1, rhAkt2, rhAkt3 (all 5 ng; lower panel), and lysates of human MCF-7, MDA-MB-123, and mouse TS1 cells (upper panel) were resolved by SDS-PAGE. Following electrophoresis, recombinant proteins and lysates were transferred to an Immobilon-P membrane and immunoblotted with 0.2 μg/mL anti-pan Akt, as described in Protocols for Immunoblotting. A fifteen second exposure to film is shown.
