Magnet Assisted Transfection (MATra) is a new, easy-to-handle, very fast and highly efficient technology to transfect cells in culture. All types of nucleic acids from plasmid DNA or siRNA to oligonucleotides can be used with the MATra approach. Data from a variety of species using cell lines or primary tissue culture have accumulated including human, monkey, mouse, rat, xenopus, pig, cat or fish. Using this new technique, nucleic acids are in a first step associated with specific magnetic nanoparticles (MagTag™).

Images: Endocytosis of NCAM in cortical neurons. After transfection membrane-localized NCAM (not endocytosed) was detected using a Cy3-coupled secondary antibody (red). Afterwards, the internalised, endocytosed NCAM was stained by a Cy2-coupled secondary antibody (green, see arrows) in the cell soma (left) and in axonal vesicles (right).

Exploiting magnetic force the full nucleic acid dose is then drawn towards and delivered into the target cells leading to efficient transfection without disturbing the membrane architecture, without causing chromosomal damage or leaving a hole in the cell membrane like other transfection technologies.

Comprehensive Protocol for All MATra-Three different procedures according to different initial situations can be followed to apply MATra:
  • MATra for adherent cells Magnet Assisted Transfection for adherent cells. The nucleic acid has to be combined with MATra-A (plasmid DNA) or MATra-si (siRNA) Reagent and MATra can be performed directly .
  • MATra for suspension cells-Magnet Assisted Transfection for suspension cells. Suspension cells have to be made adherent first by incubating them with the magnetic reagent MATra-S Immobilizer or by using polylysine plates. Then MATra-A or MATra-si Reagent loaded with the nucleic acid can be applied and MATra can be performed.
  • MATra Lipofection for adherent and suspension cells-Transfection with common lipid based (e.g. i-Fect ™, pn-Fect™ , IBAfect , Lipofectamine or Fugene) or polycationic reagents (e.g. ExGen500 or Superfect) can be enhanced by magnetic assistance. In this case, the nucleic acid is combined with MA Lipofection Enhancer in the presence of the common lipofection reagent. In addition, the MA Lipofection Enhancer can be used for viral transfection.
