HES-1 (Hairy and Enhancer of Split-1) is a transcriptional repressor that is a target of notch signaling. It contains a basic helix-loop-helix (bHLH) DNA-binding domain, an Orange domain and a C-terminal tetrapeptide WRPW motif that binds to the Groucho (Gro)/TLE/Grg family of corepressors. HES-1 can form both homo- and heterodimers with other HES family members. Dimerization is mediated through both the bHLH and the downstream Orange domain. Over the sequence used for immunization, human HES-1 shares 97% and 98% amino acid sequence identity with mouse and rat HES-1, respectively. Hes1 deficient embryos show severe defects in neuronal development, as well as pancreatic hypoplasia. It is believed to exert influence on the development of radial Glial Cells. HES-1 could play a role as a tumor suppressor in epithelial cells, and as a mediator of 17beta-estradiols have a role in proliferating breast cancer cells.
