Immunogen: Recombinant vimentin purified from E. Coli

Description/Data:
Picture

Human vimentin is a 53,521 dalton protein (466 amino acids) expressed in many cell types of mesodermal origin, and is one of the intermediate filament types that form the cytoskeleton. In the embryonic nervous system, however, vimentin is expressed by migrating neural crest cells forming the PNS, and by neural stem cells of the CNS. In some forms of glioblastoma, vimentin is re-expressed by the tumor cells and serves as a general indicator of their undifferentiated state.

Immunohistochemical staining for Vimentin is also characteristic of sarcomas (of neural, muscle and fibroblast origin) compared to carcinomas which are generally negative. Melanomas, lymphomas and vascular tumors may all stain for Vimentin. Vimentin antibodies are thus of value in the differential diagnosis of undifferentiated neoplasms and malignant tumors.

Image: hN2 Primary Neurons stained with our chicken polyclonal antibody to Vimentin, in red. Islands of Hn2 cells form after 4 days in culture forming beautiful flower like structures. Vimentin is a well established marker of early differentiating neuronal lineage cells. Taken with a 10X objective lens. Blue staining is the nuclear DNA. Protocols on data-sheet.

Images: Western blot of crude extract of human embryonic kidney stained with Vimentin, showing a single strong clean band at ~50kDa and  view of mixed neuron/glial cultures stained with Vimentin (green) rabbit antibody to GFAP antibody (red). Vimentin is expressed alone in fibroblastic and endothelial cells, which are the flattened cells in the middle of the imate which appear green. Astrocytes may express primarily GFAP, or GFAP and vimentin, and so appear red (GFAP only) or golden yellow (GFAP and Vimentin). In cells which express both GFAP and vimentin, the two protein assemble to produce heteropolymer filaments. Protocols on data-sheet.
