...Anatomical verification: In many cases, 0.4% biocytin was added to the internal solution to label the neurons following the protocol of Scott et al. (2005). Slices were fixed in approximately 0.5 ml phosphate buffered saline solution (PBS) containing 4% paraformaldehyde (PFA) following recording sessions in individual wells of a 24 well culture plate. Fixed slices were washed in PBS after fixation periods of 24 hrs to 2 weeks. Membranes were permeabilized with overnight rinse in 3% Triton X-100 in PBS. Tissue was washed in a blocking solution of PBS containing 3% Triton X-100, 1% Bovine Serum Albumin (BSA), and 0.1% saponin. MAP2 protein was immunolabeled with a MAP2 antibody (1:1000 in blocking solution, Neuromics CH22103). Biocytin was labeled with Streptavidin conjugated to Cy3 (1:500 in blocking solution, Dianova #016-160-084). Counterstaining was achieved with Ch-A647 (1:1000 Dianova) and Neurotrace green (1:100 in blocking solution, Invitrogen N21480), followed by repeated washes in blocking solution (15 min), then 3x 10 min in PBS. Slices were mounted on gelatin coated slides with fluorescent mounting medium (Vectashield H-1000, Vector labs) before confocal imaging...

 After 7 d in vitro (DIV), the cells were fixed with ice-cold (–20°C) methanol and processed for immunocytochemistry. After the appropriate serum block, the cells were incubated with primary antibodies overnight at 4°C. Secondary antibodies were applied for 3 h at room temperature. The following antibody, diluted in 1% BSA, was used: chicken microtubule-associated protein 2 (MAP2) (1:20,000; Neuromics) and anti-chicken FITC (1:100; Jackson ImmunoResearch Laboratories). Coverslips were mounted with Vectashield fluorescent mounting medium with 4`,6-diamidino-2-phenylindole (DAPI).
