Mice were perfused with 4% paraformaldehyde in PBS. Dissected brains were fixed in the same solution for 2 h at 4°C and then in 30% sacharose in PBS. Next, 40 μm free-floating sections were obtained on a cryostat. The sections were incubated in blocking solution (1% BSA and 0.3% Triton X-100 in PBS) and then incubated overnight at room temperature with the following primary antibodies: mouse anti-DARPP-32 (BD Biosciences, cat. No. 611520; 1:1000) and rabbit anti-ppEnk(Neuromics, cat. No. 14124; 1:1000). Subsequently, the sections were rinsed 5 times in PBS and incubated with the following secondary antibodies: donkey anti-mouse Alexa Fluor 488 (1:1000) and donkey anti-rabbit Alexa Fluor 647 (1:1000) for 2 h. Then, the sections were washed 3 times in PBS and mounted onto glass slides. No additional antibodies were used for tdTomato imaging....
