Contactins mediate cell surface interactions during the development of the nervous system. Contactin 1 plays a role in the formation of paranodal axo-glial junctions in myelinated peripheral nerves and in the signalling between axons and myelinating glial cells. Contactin 1 also acts as an activating ligand for Notch1, leading to generation of oligodendrocytes. Its interaction with TNR induces neuronal repulsion and an inhibition of neural outgrowth. Contactin 1 shares the L2/HNK carbohydrate epitope with L1, N-CAM, MAG and other cell adhesion molecules from nervous tissue.
