Myostatin (GDF8) is expressed uniquely in human skeletal muscle as a 12 kDa mature glycoprotein consisting of 113 amino acid residues and secreted into plasma. Myostatin is a member of the transforming growth factor ß superfamily of secreted growth and differentiation factors that is essential for proper regulation of skeletal muscle mass. Studies have shown that myostatin could play an important role in cardiac development and physiology.

Figure 2: To measure the ability of the antibody to neutralize the bioactivity of mouse GDF-8, rmGDF-8 was incubated with various concentrations of the antibody for 1 hour at 37° C in a 96 well plate. Following this preincubation period, K562 cells were added. The assay mixture in a total volume of 200 μL per well, containing antibody at the concentrations indicated, rmGDF-8 at 30 ng/mL, and cells at 2.5 x 104 cells/mL, was incubated at 37° C for 4 days in a humidified CO2 incubator. At the end of incubation, the hemoglobin level in cell lysate was measured by its psuedoperoxidase activity. The ND50 of the antibody is approximately 8 - 24 μg/mL.
