Cav1.3 Ca2+ channel or CACN4 encodes an alpha-1 subunit of a voltage-dependent calcium channel. Calcium channels mediate the influx of calcium ions into the cell upon membrane polarization. Cav1.3 voltage-gated L-type calcium channels (LTCCs) translate sound-induced depolarization into neurotransmitter release in auditory hair cells and control diastolic depolarization.
These channels also play a major role in the generation of cardiac pacemaker activity by contributing to diastolic depolarization in SAN pacemaker cells. Genetic knockout or pharmacological inhibition of Cav1.3 reverts this dependency back to HCN and protects these neurons in mouse models of Parkinson’s disease in adult mice.
