Oct-3/4 is a 34 kDa POU transcription factor that is expressed in embryonic stem (ES) cells and germ cells, and its expression is required to sustain cell self-renewal and pluripotency. Oct-3/4 is the most recognized marker for totipotent ES cells.

Oct-3/4 in Embryonic Stem Cells. Cells were cultured with recombinant human FGF-2, basic (PR27013) in the presence (L) or absence (R) of Mouse Embryonic Fibroblast-conditioned medium. SSEA-4 and Oct-3/4 were detected using anti-human SSEA-4 antibody and anti-human Oct-3/4 antibody. Cells were stained with Alexa Fluor® 568-conjugated,anti-mouse secondary antibody (SSEA-4; red) and Alexa Fluor 488-conjugated anti-goat secondary antibody. (Oct-3/4; green). Cells were counterstained with DAPI (blue).
