SMAD1 is a member of the Mothers Against Dpp (MAD)-related family of proteins. So far, eight SMADs have been identified and can be divided into 3 subgroups based on their structure and function; pathway-restricted, common mediator and inhibitory. SMAD1, 5 and 8 serve as pathway-restricted SMADs for the bone morphogenetic protein (BMP) signaling pathway. Smad1 is phosphorylated resulting in its association with the common-mediator subunit, Smad4. This heteromeric complex then translocates into the nucleus to exert transcriptional comodulator activity. |

Detection of Smad1. Lysates from 5 x 10^5 human HepG2, MDA-MB-468, and HT29 cells were resolved by SDS-PAGE, transferred to Immobilon-P membrane, and immunoblotted with 0.5 μg/mL goat anti-Smad1 as described in Protocols for Immunoblotting. A 30 second exposure to film is shown.
