Rat Oligodendrocytes were cultured on Lamnin+Fibronectin for 2 days and then differentiated with T3 for 2 days. Cells were rinsed with PBS, fixed with 4% PFA for 10 min, and then rinsed in PBS. For IHC, cells were blocked in 5% NDS in PBSTx 0.1%, incubated in MBP at 1:500 (CH22112) 4C O/N, washed 3x in PBS, incubated in secondary 1:1000 2h RT, washed 3x and then mounted. Shows rat mixed neuron/glial cultures stained with chicken Myelin Basic Protein (green). Blue is a DNA stain. Note that the Myelin Basic Protein antibody stains an oligodendrocyte and some membrane shed from this cell. Other cells in the field include neurons, astrocytes, microglia and fibroblasts, all of which are completely negative. specifically, and that the staining is concentrated in the cell bodies.
