IGF1R binds insulin-like growth factor 1 (IGF1) insulin-like growth factor 1 (IGF2). IGF1R is a tetramer composed of 2 alpha and 2 beta chains linked by disulfide bonds. The alpha chains is involved with the formation of the ligand-binding domain, and the beta chain carries the kinase domain.At a protein level is expressed in many tissues including muscle, heart, kidney, adipose tissue, skeletal muscle, fibroblasts, spleen and placenta. IGF1R is overexpressed in malignant tissues where it plays an anti-apoptotic function.

Immunohistochemical detection of IGF1R in islet in mouse pancreas. Tissue was fixed with 4% formaldehyde and cut into 10 μm thick cryostat sections. Tissue was incubated with rabbit polyclonal antibody to IGF1R at 10 μg/mL overnight at 4°C followed by incubation with Donkey anti-rabbit FITC conjugated secondary antibodies at 1:200 dilution.
